Retinal Transplants Ready for Human Trial

A major cause of hereditary human blindness, Retinitis Pigmentosa (RP), has been resolved in rat trials by researchers at the RIKEN Center for Biosystems Dynamics Research (BDR) in Japan. Degenerated rat retinas received transplants and removal of grafted cells, which resulted in improved connections to host retinas and better light-responsiveness.

Study leader Michiko Mandai said, “We have previously reported work using mouse embryonic- and induced pluripotent stem cell – (iPES)-derived retinas in a mouse model of retinal degeneration.”

However, the deputy project leader in the Laboratory for Retinal Regeneration at RIKEN BDR said, “As far as we know, this is the first report of the timed removal of particular grafted cells being shown to improve retinal function in an animal model using tissue from human stem cells.”

Since the retinal sheets were created from human stem cells, the novel feat can now be tested in human clinical trials, according to the study authors in the January 21, 2022 edition of iScience.

“Since retinal ganglion cells form optic nerves to send the signal to the brain, a higher light responsiveness could theoretically lead to improved sensitivity to light or better resolution in visual function. Genetic modification in human stem cell-derived retinas showed a substantial functional improvement [in light responsiveness], compared with wild-type graft retinas. Depending on the retinal network of the transplanted area, light responsiveness of retinal ganglion cells may be correlated with increased signaling to the brain, so could lead to an increased light sensitivity or better resolution. Additionally, we were able to observe detailed host-graft synapse formation in the absence of graft bipolar cells, which previously was difficult. A frequently asked question in retinal cell therapies concerns whether retinal ganglion cell responses are really originated from the graft photoreceptors. The presence of multiple synaptic markers at the host-graft cell contact site suggests formation of some synapses between these cells and indicates that graft photoreceptor light responses may well be transmitted to host retinal cells. We can now move forward to applying this strategy in clinical studies, which we expect to improve clinical outcomes and be generally useful in stem cell-based therapies targeting retinal degeneration. If the clinical grade ISL1-deleted stem cell-derived retinas can first pass extensive safety testing, including for tumorgenicity, we estimate that this may take 4-5 years. While we are currently targeting end-stage disease eyes with stem cell based therapy, we think this strategy could also be applied to those with remaining central vision, to help increase the pericentral visual field or delay disease progression,” Mandai said.

Contaminated Water Detected by Smartphone Screens

Billions of smartphones and tablets worldwide could be used to sense toxins in water and soil by way of the touchscreen technology used in everyday practice without any modifications. The report was published in the journal Sensors and Actuators B.

Researchers at the University of Cambridge have successfully shown how a regular touchscreen can identify contaminants by dropping liquid samples on the screen. Since touchscreen sensitivity is comparable to lab-based equipment, it can be used in unsterilized environments.

For now, this novel accomplishment is a proof of concept, but it can be expanded for various applications such as biosensing or medical diagnostics. Smartphones interpret electrical fields when a finger disrupts the electrical field running through a touchscreen.

Dr. Ronan Daly from Cambridge’s Institute of Manufacturing, who co-led the research, said, “We wanted to know if we could interact with the technology in a different way, without having to fundamentally change the screen. Instead of interpreting a signal from your finger, what if we could get a touchscreen to read electrolytes, since these ions also interact with the electric fields?”

During trials, researchers pipetted several liquids onto smartphone screens. Depending on the concentration of ions and their charge, the measurements were recorded from each droplet.

The new technology could be used to detect arsenic contamination in drinking water. Arsenic is a common contaminant found in groundwater but is filtered out by most municipal water systems. Arsenic water contamination is a substantial problem in parts of the world without water treatment.

“In theory, you could add a drop of water to your phone before you drink it, in order to check that it’s safe. For example, if we could get the sensitivity to a point where the touchscreen could detect heavy metals, it could be used to test for things like lead in drinking water. We also hope in the future to deliver sensors for home health monitoring,” said Daly.

Novel ‘Cure’ For Cancer Treatment

A new report published in Nature titled “Decade-long leukemia remissions with persistence of CD4+ CAR T cells” highlights a novel treatment called CAR T, which is being hailed as a “cure” for cancer by Dr. Carl June, the principal investigator for the trial at the University of Pennsylvania.

June said, “Now we can finally say the word ‘cure’ with CAR T cells.” In 2010, when the trial began, the idea for this sort of therapy “was way out there,” he added.

June and his colleagues reported that the CAR T therapy wiped out cancer in two out of the three initial early-trial patients, all of whom had chronic lymphocytic leukemia. Though the cancer disappeared, the CAR T cells stayed circulating through patients’ bloodstreams, almost guarding the body against future attacks.

In CAR T therapy, T cells (white blood cells that fight viruses) are removed from a patient’s blood and re-introduced after being genetically engineered to fight cancer. Patients’ T cells are taught to recognize and destroy B cells, a key player in cancer formation throughout the body. Without B cells – or cancer – patients require regularly-scheduled infusions of immunoglobulin infusions, which replenish antibodies.

Many people with blood cancer have benefitted from CAR T therapy, which has proved especially effective against acute leukemia. Roughly a third of patients who underwent the CAR T therapy have gone into remission, but many patients later relapse.

Dr. John F. DiPersio, chief of the division of oncology at Washington University in St. Louis, said, “The question is not only why some patients relapse or are resistant to therapy but why are some patients cured?”

He continued, “Oncologists don’t use words like ‘cure’ lightly or easily or, frankly, very often,” he said. “I guarantee that it’s not being used lightly. The patients we treated had far advanced disease,” he noted, adding, “the biggest disappointment is that it doesn’t work all the time.”

Winter Storm Pool Recovery

Adverse winter weather can affect many aspects of our lives, even the pool you might have forgotten about during the cold season. Snowstorms and heavy frozen rain can impact your pool in many ways. Read on to learn how and what to do if winter weather brings heavy snow, ice, and high winds to your backyard.  

Heavy Snow
Even with a mesh cover on your pool, excessive snowfall can be an issue. Until the snow can melt, it cannot pass through the cover, which means it accumulates. Solid covers trap the snow (even if it melts) until you can remove it with a cover pump. When heavy snow falls and piles up over five inches, it can damage your pool cover. The excess weight alone can put a lot of stress on your cover, which could rip or fail if it breaks. If the snow is light and fluffy, you can use a leaf blower to push it off the pool cover. If the snow is heavy or wet, instead, use a soft push broom, leaf rake, or telescoping pool pole to get the snow off. Never step onto the pool cover to remove snow. Remove snow from the edges, with your feet on solid ground, not ice. 

Ice
Freezing snow or rain that freezes later is a challenging aspect of winter for pool owners. As the ice melts away, remember to use a cover pump to remove it. Do not attempt to crack or break the ice, as you can tear the pool cover.

Fallen Limbs
Ice storms and blizzards can throw broken tree branches and twigs on the ground. If any fall on the pool cover, remove them as carefully as possible. Check thoroughly for damage and make an urgent plan to repair or replace the pool cover as soon as possible. If debris is introduced into the pool water through a rip or hole in the cover, remove as much of it as you can to prevent a swampy mess once spring comes. Also, check the pool’s water balance to ensure there are no chemical issues.

High Winds
If not appropriately secured, pool covers can swing to and fro in high winds. Ensure your pool has adequate water tubes or blocks attached securely to the cover. Tighten winches, straps, and buckles if necessary to verify that the cables are tight.

Malicious Authenticator App Stealing Bank Information

An Android app posing to provide two-factor authentication in the Google Play store has turned out to be malicious. With over 10,000 phones affected, the app was available for 15 days.

The malicious app, called 2FA Authenticator, is no longer available on the Google Play Store. The app description read, “A secure authenticator for your online services, while also including some features missing in existing authenticator apps, like proper encryption and backups.”

In reality, the app was stealing financial information by copying an open-source code from a well-known legitimate app (Aegis Authenticator) and then copying malicious code into their own app. The 2FA Authenticator app was able to pass through the Google Play Store’s security checks but turned malicious once installed on an Android user’s phone or tablet.

Most apps ask for user permission once installed on the smartphone or device. The 2FA Authenticator app requested “critical permission,” which allowed it to execute a number of tasks such as:

  • Disabling keyboard lock
  • Disabling password security
  • Downloading third-party apps (and updates)
  • Running in the background after the app is closed
  • Placing an overlay on other apps
  • Having complete access to user data

Once the 2FA Authenticator app is installed, a Remote Access Trojan (RAT) is downloaded and installed. This trojan, called Vultur, records screens and keystrokes to capture details entered into banking apps. In return, criminals can quickly empty bank accounts, cryptocurrency wallets, and more.

How a Snow Cave Could Save Your Life

You might think it is substantially easier to find shelter in warm weather, but did you know snow caves can actually protect you in chilly, frigid temperatures? Snow caves can be excellent shelters and are generally made from deep snow, usually at the bottom of a hill.

Despite being freezing cold inside, snow caves are usually warmer than outside air temperatures, especially when considering windchill. If built correctly, a snow cave can sustain significantly warmer temperatures inside when compared to the exterior.

Snow caves can be intelligently constructed with an entrance tunnel leading to a small chamber with a domed roof. The chambers are usually big enough for two to four people and can showcase snow “benches” built into the floor.

To keep warm air in the dome, the roof is kept as low as possible, allowing it to support heavy snow weight from above. It is best to ensure the chamber is built on a slight incline compared to the entrance. When heat is trapped inside, the slight elevation helps it stay contained rather than floating down to the floor and escaping through the entrance tunnel.

Snow caves are wonderful insulators, maintaining a significantly higher temperature than outdoor temperatures, and can also prevent windburn.

Proper winter clothing is still required to maintain comfort within a snow cave. Keep survival kits with sleeping bags, gloves, blankets, ground pads (yoga mat), etc., to get the most warmth you can out of the cave.
